14 Rudenko P.A. The role of intestinal dysbacteriosis in the mechanisms of the formation and progression of surgical infection in cats

The article presents data on the role of intestinal dysbacteriosis in the mechanisms of the onset and development of accidental purulent-inflammatory processes of soft tissues in cats. It is shown that in the formation and progression of surgical infections in cats, dysbacteriosis of the intestine arises and intestinal translocation of opportunistic pathogens into the focus of purulent inflammation is evidenced by the determination of the antigenic kinship of cultures of intestinal microorganisms and wound sites according to agglutinable properties, as well as the comparison of antibioticograms of microorganisms isolated from cats from a soft tissue biopsy and fecal samples with an operating wound and with different forms of surgical infection . Depending on the severity of the surgical infection, the microbiota changes in the intestine, and in the group of animals with sepsis they approach critical values, which indicates the development of irreversible reactions in the body. Key words: dysbacteriosis, ovariohysterectomy, purulent wound, abscess, sepsis, cats.

The DNA of Chlamydia trachomatis (CT) of the new «Swedish» variant (nvCT) with a deletion of 377 b.p.  in the orf1 gene of a cryptic plasmid was detected in clinical specimens from aborted cows. All of them had diagnostic titers of specific chlamydial antibodies in compliment fixation test. Molecular omp1 typing demonstrated the complete homology of the nucleotide sequences of the bacterial strains identified with the CT genovar E (variants of the E1 subtype: E/Bour, Sweden2, Sweden3). Genetic material (DNA) of other Chlamydia species, including Chlamydia psittaci and/or Chlamydia abortus, as well as other potential causative agents of infectious abortions of livestock (Campylobacter spp., Listeria spp., Toxoplasma gondii, Salmonella spp., Brucella spp., etc.) in the specimens studied by real-time PCR commercial kits was not detected. Key words: Chlamydia trachomatis, chlamydial abortion, livestock, cattle, PCR, novel Swedish variant.

This paper presents the results of determination of optimal conditions for obtaining hemadsorption inhibiting antisera against African swine fever virus (ASFV) seroimmunotypes III and IV. A preliminary injection of a minimum protective dose of an attenuated hemadsorbing strain followed by infection with a homologous virulent ASFV reference strain together with phosphonoacetic acid (FUC) injection to reduce the probability of death from the disease was shown to be the most effective approach here. Thus, serotype III and IV antisera active in haemadsorption inhibition (HAdI) tests with titers or 1:80 to 1: 640 were obtained. Key words: African swine fever, serotype-specific sera, hemadsorption inhibition test.

45 Naumenkova V.A. Influence of collateral effects of vaccination on stallions’ semen

Vaccination being the strong stress-factor has the strong influence on the different systems of organism. The question of the influence of immunoprophylaxis of infections diseases on reproductive functions of animals, especially stallions, practically stays open. Studied investigation showed that vaccination of stallions against dermatomicose  by vaccine Polyvak-TM decreased the quality of semen, especially mobility and surviving after freezing- thawing process. It is important to take into account this fact when cryoconservation of semen. Rehabilitation of the quality of fresh semen happens not earlier than in 25 days after the first vaccination, for deconserved semen – not earlier than in 55 days. Key words: stallions, sperm, Polivak-TM vaccine, cryoconservation.

LABORATORY PRACTICE

56 Zakharychev V.V., Brevnova B.O., Kuzenkov A.V., Komaritskikh M.Yu., Trubnikova O.N. Developement of HPLC-MS/MS method for the analysis of free lipophilic biotoxins in mollusks

There are no any valid methods in Russia for the determination of lipophilic biotoxins in molluscs compliant with the European Union regulations. The fact makes great hindrance for shellfish export. We have developed reverse-phase chromatographical conditions using acidic buffer as a mobile phase and tandem mass-spectrometric detection parameters for nine biotoxins: okadaic acid (OA), dinophysis toxins 1 and 2 (DTX1 and 2), pectenotoxin 2 (PTX2), yessotoxin (YTX), homoyessotoxin (homoYTX), and azaspiracids 1 – 3 (AZA1 – 3). For OA, PTX2, YTX, AZA1 a 3-days validation procedure was performed using the Comission Decision 657/2002/EC as guideline with fifty four samples of tissues of oysters, mussels and scallops fortified with 0,5; 1; 1,5 times of the EU permitted limits of the biotoxins. Linear response in the investigated concentration range was observed; the recovery was within 97 – 108 %, the intermediate precision values were always lower than 8.5 %. The values of decision limit CCα amounted to 174 μg/kg for OA, 170 μg/kg for PTX2, 1083 μg/kg for YTX, 170 μg/kg for AZA1. The limits of detection and limits of quantitation for these four biotoxins were estimated, as well as expanded uncertainties in measurements. The procedure is highly sensitive and selective enough to determine isomeric OA and DTX2 separately. The suitability of procedure for routine analysis of lipophilic marine biotoxins was shown. Key words: azaspiracids, validation, dinophysis toxins, high performance liquid chromatography, marine biotoxins, mass-spectrometry, okadaic acid, pectenotoxins, phycotoxins, yessotoxins.
