18 Stafford V.V., Streltsova E.B., Raev S.A., Yushakov A.G., Zaberezhny A.D.Aliper T.I.  Application of immunohistochemistry method in diagnosis of porcine circovirus associated diseases

Porcine circovirus associated diseases (PCVAD) remain one of the urgent problems in pig industry. Porcine circovirus type 2 is a causative agent of the disease that has an ubiquitous distribution among pigs. In the absence of pathognomonic signs of the disease, laboratory diagnostic, in particular, immunohistochemistry (IHC) is required to establish the diagnosis – PCVAD. The present study describes the visualization of viral antigens based on development of the IHC method for detection of PCV-2 capsid protein in pathological material. As a result of this work, it was found that the developed IHC method allowed to establish a relationship between the presence of PCV-2 and pathologic observations. Key words: PCVAD, PCV-2, immunohistochemistry, monoclonal antibodies, capsid protein.

22 Vlasov M.E., Sereda A.D., Balyshev V.M.  African swine virus maintenance in stable flies (Stomoxys calcitrans) and blowflies (Calliphoridae)

The heparinized virus-containing blood of pigs, infected with the virulent strain Stavropol 01/08 of ASF virus, was being fed in the laboratory to stable flies and blowflies. As show the research results of suspensions from flies using PCR-RV, and the extraction of the agent of ASF in cell culture, virus persisted in the flies for at least 5 days. After intramuscular injection the homogenates of stable flies and blowflies, that feeding virus-containing blood, two pigs were infected on the 7th and 10th day. The disease was followed with typical symptoms and resulted in death on 12th and 15th day. Autopsies revealed pathological changes typical for this infection. The results suggest that these flies can be vectors of ASF. This is indirectly confirmed by the increase in the number of ASF outbreaks in domestic pigs and wild boars in the summer and autumn period. Key words: African swine fever, hemadsorption, flies, infectious activity, viral genome, pigs.

34 Filippova O.B., Saranchina E.F., Krasnoslobodtseva A.S.  Medicinal plant for new-calved cows

The possibility of use the wild plants Artemisia vulgaris L. and Salvia pratensis L. as a prophylactic and stimulating feed additive in the rations of newly calved cows was studied. It was found that the feeding of these phyto-additives during the first 4 – 5 weeks after calving increases the milk productivity of cows by 9,8 %, protein content in milk by 6,5 % and its density. Milk fat content corresponded to the level of metabolism, which was more balanced in the animals receiving the feed additive than in the control cows, which were given only the basic ration. The bactericidal effect of herbal supplements made it possible to exclude the occurrence of clinical mastitis and to reduce the number of somatic cells in the milk of healthy experimental animals by 27,8 % in comparison with healthy control cows. Key words: cows, mastitis, medicinal herbs, milk quality, productivity.

38 Chistiakova I.V., Kuzmina T.I., Denisenko V.Yu. The evaluation of the bovine spermatozoa viability with induced capacitation before and after thawing

It has been shown that the preliminary capacitation of bovine spermatozoa before freezing increases the number of viable spermatozoa after thawing [2]. In artificial insemination, thawed spermatozoa are transferred to the female reproductive tract, where the capacitation occurs. In this study, we examined the effect of conditions, which provide the passing of capacitation on thawed spermatozoa, preliminarily capacitated before freezing. It has been shown that the incubation of the control thawed (without prior capacitation) spermatozoa in the absence, as well as in the presence of dbcAMP and IBMX, prolactin (PRL) and GTP for 4 hours did not lead to the change in the ratio of live/dead cells. Incubation of thawed spermatozoa, which had been capacitated previously before freezing, during 4 hours with dbcAMP and IBMX, PRL and GTP, led to an increase in the number of viable spermatozoa. The usage of chlortetracycline probe (determining the location of the fluorescence of chlortetracycline in spermatozoa) showed that in intact thawed spermatozoa the ratio of cells with different functional status - uncapacitated, capacitated and acrosome-reactive, was similar among all experimental groups. In thawed spermatozoa, which had previously capacitated before freezing, the decrease in the number of capacitated and the increase in the number of acrosome-reactive cells in the presence of dbcAMP and IBMX, and also PRL and GTP were noted. Thus, the second incubation of thawed spermatozoa for 4 hours, which had previously been capacitated before freezing, did not increase the number of viable cells. Key words: capacitation, freezing, bovine spermatozoa.

56 Usadov T.R., Koltsov A.Yu., Sukher M.M., Morgunov Yu.P., Yanzhieva D.V., Salnikov N.I.  Test-system for detection of lumpy skin disease virus genome by Real-Time PCR

This article presents data on the development of a test-system for detection of lumpy skin disease (LSD) virus genome by Real-Time PCR. This technique is based on the amplification of a fragment (113 b.p.) of LSDV 129 gene using the original oligonucleotide primers and fluorescent-labeled hybridization probe. Analytical specificity of the developed test system was evaluated by testing the strains of LSD virus, sheep pox and goat pox viruses, contagious ecthyma virus, as well as biological samples from clinically healthy animals and intact cell cultures. The analytical sensitivity of the developed test-system was determined using tenfold serial dilutions cultural virus contained material, was 1,0±0,15 lg TCID50/ml. To create a positive control of amplification, the fragment of LSDV 129 gene synthesized during Real-Time PCR was cloned as part of the plasmid vector pGEM-T Easy in the E. coli bacterial cells. It was established that the range of plasmid DNA concentration, at which the amplification linearity is observed, is from 3,0х109 до 3,0 DNA copies/μl. Key-words: lumpy skin disease, virus, test-system, Real-Time PCR.

Efficiency and detection limit (LOD) of the method were calculated, sensitivity and specificity of oligonucleotides were analyzed. To ensure the reproducibility of the study, two operators were conducted independently using different equipment of the same brand at different times. Positive control samples were developed. Proven a direct correlation of the threshold cycle Ct from the logarithm of the concentration of the DNA matrix, which is in the required range of measurements (R2≥0,98), which is also confirmed by the obtained values of the slope of the calibration curve (slope= –3,4; –3,4 and is –3,2). As a result of research it is shown that the analytical sensitivity of PCR is high (90, 70 and 30 copies of the target DNA, respectively), LOD is not more than 0,1 %, which meets the criteria. Specificity of oligonucleotides was checked on the panel of positive and negative samples. Within the working range of the method of false-positive and false-negative results was not revealed, indicating a high accuracy of the reaction. Key words: validation, viral diarrhea virus, the efficiency of reaction, limit of detection (LOD) techniques, the sensitivity and specificity of the work of oligonucleotides.
